Located in the town of Penzance, the station is the western terminus of the 491km Cornish Main Line from London Paddington station. It is also the western terminus of the Night Riviera sleeper train to and from Paddington.The main train shed contains platforms Platforms 1, 2 and 3; Platform 4 is on the south side in the open. One side of the station is bordered by the sea wall near the harbour, whereas the other side is carved into the hillside.

Both the southernmost station on the National Rail network and the westernmost station in England, Penzance Station is not the most westerly station in all of Great Britain because Scotland has stations further west. The station is run by First Great Western. Penzance.
